Category Of Drug

Broad-spectrum bacteriolytic, ß-lactam antibiotic in the penicillin family

Side Effects

Body as a whole - Fungal infections, severe allergic reactions, joint pain, or general ill feeling, severe tingling, sleeplessness, numbness and pain. 

   Gastrointestinal - Nausea, vomiting, diarrhea, antibiotic-associated colitis, black hairy tongue and intestinal bleeding. 

   Hypersensitivity - Anaphylactic shock, redness of skin, skin inflammation, hives, inflammation of blood vessels, itching, severe blistering and peeling. 

   Liver - Jaundice and liver inflammation. 

   Genitourinary - Crystals in urine, dark colored urine and vaginal itching or discharge. 

   Blood - Anemia, decrease in white blood cells, platelets, increase in eosinophils and bruising. 

   Central Nervous System - Reversible hyperactivity, agitation, anxiety, sleeplessness, confusion, convulsions, behavioral changes and dizziness. 

   Miscellaneous - Tooth discoloration, hear burn, white patches or sores inside mouth or on lips, easy bruising and bleeding.

Drug Dose Information

Oral
Adult- 250 mg every 8 h, double in severe infection.
Otitis media: 1g every 8 h.

Enteric fever: 2 to 4g daily in divided doses for 14 to 21 days.

Intramuscular injection 500 mg every 8 h.

Intravenous injection or infusion: 500 mg every 8 h, increase to 1g every 6 h in case of severe infection.
Child up to 10 years- 125 mg every 8 h, double in severe infections.

Otitis media: 40 mg/kg body weight daily in three divided doses.

Enteric fever: 50 to 100 mg/kg body weight in three divided doses for 14 to 21 days.

Intramuscular injection: 50 to 100 mg/kg body weight in divided
doses.

Intravenous injection or infusion: 50 to 100 mg/kg body weight in divided doses.

Amoxicillin is available as a tablet, capsule, chewable tablet, syrup and pediatric drops taken orally. It is usually taken every 12 hours (twice a day) or every 8 hours (three times a day) with or without food. It is recommended that at least 10 days' treatment for any infection caused by Streptococcus pyogenes to prevent the occurrence of acute rheumatic fever.

Storage

Tablet, Capsule and Oral suspension: Store protected from moisture at a temperature not exceeding 30°C. Injection: Store protected from moisture in a sterile, tamper evident container sealed so as to exclude micro-organisms at temperature not exceeding 30°C.

Contra Indications

Hypersensitivity to penicillins.

Malaria : Malaria is a mosquito-borne disease caused by a parasite. People with malaria often experience fever, chills, and flu-like illness. Left untreated, they may develop severe complications and die. In 2010 an estimated 219 million cases of malaria occurred worldwide and 660,000 people died, most (91%) in the African Region Quinine derivatives /Quinine sulphate: Destroys the asexual forms of Plasmodium in three days. Used in the treatment of cerebral malaria, it clears the clogs in the brain capillaries caused by Plasmodium falciparum. Not toxic in recommended dosage but excessive dosage may cause temporary deafness. Pregnant women should not be given quinine derivatives. Acridine derivatives/Mepacrine and Quinacrine: Destroys the asexual forms of plasmodium after the third or fourth day. Toxic effects are temporary yellow coloration of skin, face, eyes and urine. Biguanie/Paludrine: Destroys the tissue forms and can be used as a prophylactic drug. - Prophylactic dosage – 300 mgs once a week - For treatment – 300 mgs daily for 5 to 10 days

Interactions

Increased levels with disulfiram and probenecid. Decreased effects with tetracyclines and chloramphenicol. Potentially Fatal: Increase effects of oral anticoagulants.
