Side Effects

CNS- Headache, dizziness, drowsiness, aggression, agitation, depression, hallucination. 

 Itching, hair loss, severe allergic reactions, photosensitivity, blistering. 

Eye and ENT- Blurred vision. 

Gastrointestinal- Flatulence, abdominal pain, stomach upset, nausea, diarrhea, dry mouth, constipation, GI fungal infection, inflammation of pancreas and intestine, mouth ulcer, taste disturbance. 

Genitourinary - Breast enlargement in men, kidney disorder. 

Liver- Occurrence of confusion, altered level of consciousness and coma as a result of liver failure(Hepatic encephalopathy) , liver failure, inflammation of liver with or without jaundice. 

Blood- Decrease in white blood cells. 

Local- Injection-site reactions including mild local redness and itching with IV administration. 

Musculoskeletal- Muscular weakness, muscle pain. 

Respiratory- Asthma, respiratory tract infection.

Drug Dose Information

Oral
Orosive Oesophagitis
Adult: 40 mg once daily for 4 week, extended for another 4 week if necessary. Maintenance: 20 mg once daily.

Child: <20 kg: 10 mg once daily for 8 wk; =20 kg: 10-20 mg once daily for 8 week. 

Hepatic impairment: Severe impairment (Child-Pugh class C): Not more than 20 mg daily.

Oral :
Peptic ulcer
Adult: 20 mg bid for 7 days or 40 mg once daily for 10 days given as a triple therapy with amoxicillin and clarithromycin.

Oral:
NSAID-Associated ulcer
Adult: 20 mg once daily for 4-8 week.

Oral:
Zollinger-Ellison syndrome
Adult: Initially, 40 mg twice daily. Usual range: 80-160 mg daily. Doses >80 mg should be given in 2 divided doses.

How To Take

It available as a tablet or capsule to taken orally, once a day, one hour before meal.

Injection: Powder for injection: Store at (20 - 25°C). Protect from light. Reconstituted solution for injection should be stored at room temperature up to 30°C and administered within 12 hr after preparation.

Contra Indications

Hypersensitivity. It should not be co-administered with Methotrexate, clopidogrel and atazanavir.

Bronchitis : Bronchitis is an inflammation of the mucous membranes of the bronchi (the larger and medium-sized airways that carry airflow from the trachea into the more distal parts of the lung parenchyma) Treatment Conventional treatment for acute bronchitis may consist of simple measures such as getting plenty of rest, drinking lots of fluids, avoiding smoke and fumes, and possibly getting a prescription for an inhaled bronchodilator and/or cough syrup. In some cases of chronic bronchitis, oral steroids to reduce inflammation and/or supplemental oxygen may be necessary.

Interactions

May interfere with the elimination of drugs metabolised by CYP2C19. Changes in gastric pH can affect the bioavailability of ketoconazole and Fe salts. Clarithromycin and amoxicillin may increase plasma level. May cause changes to prothrombin time when used with warfarin. Avoid concurrent use with clopidogrel.
